RNA polymerase

Copies selected DNA instructions into RNA.

LIBRARY BASIS · BACTERIAL ACCOUNT

The question behind this machine

The library emphasizes the reciprocal dependence between gene expression and the proteins that carry it out. Its thread also discusses eukaryotic transcription. This article uses bacterial core polymerase and sigma-factor organization rather than importing Pol II, a nucleus or eukaryotic general transcription factors.L1

A template and a physical reader

The Thermus thermophilus holoenzyme structure resolves a multisubunit bacterial RNA polymerase associated with an initiation factor. This provides a physical model for how a core catalytic assembly and sigma cooperate during transcription initiation. Transcription makes RNA from a DNA template; it does not translate that message directly into protein.P1

From inherited information to an assembled enzyme

A functioning cell must express the relevant polymerase chains and assemble them in a compatible form. The library’s recurrence argument points out that those gene products are themselves made through gene expression. This is a systems-level dependency, not evidence that one isolated polymerase recreates the entire expression system.

Recognition, catalysis and movement

Useful activity requires a productive template arrangement, selection of incoming nucleotides and coordination of chain extension with movement. Promoter recognition and elongation are different tasks, so an initiation defect need not indicate failure of the same chemical step as an elongation defect. The exact kinetics and factor requirements are organism- and condition-dependent.

Checking the origin inference

The origin target must be specified: contemporary bacterial transcription, a simpler ancestral polymerase or the first template-copying chemistry. Homology and experimentally useful alternative architectures are relevant to that comparison. The modern recurrence of expression machinery supports the author’s integration argument, while leaving the historical ordering and possible predecessors open to investigation.
