FEATURED MACHINE 07 / 12

Sec protein-translocation machinery

Moves selected proteins through the cell membrane.

LIBRARY BASIS · BACTERIAL ACCOUNT

The question behind this machine

Three library threads connect targeting signals, membrane insertion and secretion. Their common question is how a newly made protein reaches a compatible location. The bacterial account distinguishes the targeting system from the channel and from the energy source that moves a substrate.L1 L2 L3

A route through the membrane

The bacterial SecA–SecY complex structure links a cytoplasmic ATPase to a membrane channel. It supports a physical account of how a substrate-translocation pathway can be coupled to an energy-using partner. A channel capable of passing an unfolded chain must also remain compatible with a functional membrane boundary.P1

Targeting is a separate recognition problem

Structural work on signal recognition particle and its receptor illuminates a targeting interaction that precedes productive membrane engagement for suitable substrates. This does not make every exported protein an SRP substrate, nor does it show that all Sec transport is powered through the same route. Address recognition, delivery and translocation should be described separately.P2

Insertion, assembly and sidedness

Membrane-protein production includes more than passing a chain through a hole. Appropriate topology, interactions with lipids and assembly with partner chains can affect the final outcome. The library’s discussion of Sec and YidC is useful context, while each proposed Aquifex component and substrate relationship still needs organism-specific support.

Quality handling and origins

A protein in the wrong location, one that fails to insert and one that inserts but misfolds pose different problems. The historical comparison should consider simpler membrane insertion, alternative targeting and selectable partial functions. The modern match between a signal and its reader contributes to the design argument, but a signal alone does not demonstrate a complete delivery system.
